TY - JOUR AU - ., Kisman PY - 2017 TI - POLA PERTUMBUHAN AWAL TANAMAN KEDELAI PADA KONDISI CEKAMAN INTENSITAS CAHAYA RENDAH DAN PEMBERIAN INHIBITOR PLASTIDA (UJI CEPAT TOLERANSI KEDELAI TERHADAP CEKAMAN NAUNGAN JF - CROP AGRO, Scientific Journal of Agronomy; Vol 1 No 2 (2008): Jurnal Crop Agro KW - N2 - ABSTRAK Penelitian ini bertujuan untuk mengetahui pola pertumbuhan awal tanaman kedelai pada kondisi cekaman intensitas cahaya rendah dan pemberian inhibitor plastida. Percobaan disusun berdasarkan rancangan acak lengkap pola faktorial dengan tiga faktor: faktor genotipe (G 1 = genotipe toleran, varietas Ceneng; G 2 = genotipe peka, varietas Godek), faktor jenis inhibitor plastida (I 1 = kontrol/aquades, I 2 = lincomycin , dan I 3 = chloramphenicol ), dan faktor cahaya (L 1 = terang total dan L 2 = gelap total). Dengan demikian terdapat 12 kombinasi perlakuan dan masing-masing perlakuan diulang tiga kali. Hasil penelitian menunjukkan bahwa: (i) Benih kedelai yang ditumbuhkan pada kondisi cahaya penuh menunjukkan perkembangan awal mengikuti pola fotomorfogenesis dengan ciri-ciri kandungan klorofil a dan b yang lebih tinggi, kotiledon dan hipokotil berwana hijau, tumbuh bulu akar yang lebih banyak, hipokotilnya lebih pendek dan kuat, posisi kotiledon tegak dan kotiledon berkembang dengan baik dan terbuka. Sebaliknya, perkembangan kecambah kedelai pada kondisi gelap total mengikuti pola skotomorfogenesis dengan cirri-ciri kandungan klorofil a dan b rendah, kotiledon dan hipokotil berwana pucat, tidak tumbuh bulu akar, hipokotilnya lebih panjang dan ramping, posisi kotiledon mernduk dan kotiledon tidak berkembang dengan baik dan masih tertutup. (ii) Pemberian inhibitor translasi plastida ( lincomycin dan chloramphenicol ) dapat menghambat pembentukan klorofil (khususnya kolorofil a ), keduanya tidak memberikan penghambatan yang berbeda. (iii) Kedelai toleran dan peka naungan tidak menunjukkan pola perkembangan awal yang berbeda pada kondisi cekaman cahaya dan jenis inhibitor plastida. ABSTRACK Aim of the study was to know the seedling growth pattern of soybean under the effect of low light intensity stress and plastid inhibitors. The study was conducted using factorial in completely randomized design (CRD) with three factors: soybean genotypes (G1 = tolerant genotype, Ceneng; G2 = sensitive genotype, Godek); plastid inhibitor ( I 1 = control/aquadest, I 2 = lincomycin, I 3 = chloramphenicol); and light (L1 = light, L2 = dark), each combination was in three replicates. Results of the study showed that: (i) seedling growth pattern of soybean germinated under fully light was in photomorphogenesis as indicated by higher contents of chlorophyll a and b, green color of cotyledons and hypocotyls, more root hairs, shorter and stronger hypocotyls, no apical hock, and well developed and opened cotyledons. Contrastly, under dark condition, growth pattern of soybean seedling was in scotomorphogenesis as indicated by lower contents of chlorophyll a and b, pale color of cotyledons and hypocotyls, less root hairs, longer and weaker hypocotyls, along with apical hock, and cotyledons were not develop and open. (ii) Application of plastid inhibitors (lincomycin and chloramphenicol) prevented the formation of chlorophyll (especially for chl a) though no significant different on both. (iii) No difference between tolerant genotype and sensitive genotypes of soybean on seedling growth pattern under low light intensity stress and the application of plastid inhibitors.
